The striker is now a free agent after being handed his clearance the past weekend.
Macuphu joined Royal AM in 2022 having been recruited from TS Galaxy.
He has played for Jomo Cosmos, Chippa United, Golden Arrows, SuperSport United, and Bidvest Wits, amongst teams in the PSL.
Thwihli Thwahla general manager Richard Makhoba has confirmed Macuphu has been served with his clearance certificate and is now free to join a team of his choice.
"We parted ways amicably," confirms Makhoba, speaking to KickOff.com.
"There was tough competition up front amongst the strikers. He felt that he was not getting much game-time and wanted to continue his career elsewhere. We understand his decision, taking also into consideration his age.
"He is a true professional both on and off the field. He is a good player and we thank him for his contribution to the club," added the club official.
Royal AM remain banned by FIFA from registering players due to outstanding payments due to their former marksman Samir Nurkovic.
Makhoba assured their fans that only Macuphu has left the club, over and above the exit of long-serving Hugo Nyame.
